FIELD: medicine.
SUBSTANCE: invention refers to biotechnology and can be used for the recombinant production of human coagulation factor VIII (FVIII) with deleted B-domain. The procedure involves constructing an expression plasmid for recombinant human FVIII with deleted B-domain in dihydrofolate reductase (DHFR) gene knockout CHO cells; the above plasmid contains a region of origin of replication of the pUC plasmid, a beta-lactamase (bla) open reading frame and a bla gene prokaryotic promoter, a human Epstein-Barr virus terminal repeat (EBVTR) site, functional elements of Chinese hamster elongation factor 1 alpha gene, Kozak sequence; an open reading frame of a human FVIII gene with deleted B-domain; an internal ribosomal entry site (IRES) of EMCV virus; an open reading frame of DHFR expressed as a part of bicystronic mRNA with the gene coding human FVIII gene with deleted B-domain (IRES DHFR). The produced plasmid is used to transform a CHO cell to produce a factor producer cell.
EFFECT: invention enables substantially increasing the production effectiveness in the CHO cells and cutting the expenses for producing the human recombinant FVIII with deleted B-domain of pharmaceutical importance.
