FIELD: biotechnology, biochemistry, enzymes.
SUBSTANCE: invention relates to a method for preparing immobilized biocatalysts based on enzymes included into gel carrier matrix that are able to carry out enzymatic transformation of corresponding substrates in periodic and continuous regimens. The claimed biocatalyst is insoluble enzyme preparation or enzymes preparation included into matrix of polymeric gel based on polyvinyl alcohol. Insoluble enzyme preparation or enzymes preparation represents cross-linked gel, or cross-linked coagulate, or cross-linked protein crystals, or particles consisting of enzymatically inactive material to which enzyme(s) are cross-linked covalently. Matrix of polymeric gel represents polyvinyl alcohol cryogel of the following ratio of the parent components, wt.-%: insoluble enzyme preparation, 1-20; polyvinyl alcohol, 8-16; water, up to 100. Method for preparing the claimed biocatalyst involves dispersing insoluble enzyme preparation in polyvinyl alcohol an aqueous solution followed by freezing the prepared dispersion at (-5)-(-40) C for 4-48 h and further defrosting at the rate 0.001-0.2 C/min. Invention provides preparing active, stable and mechanically strength biocatalysts comprising much more amount of immobilized enzyme as compared with analogues prototype (up to 680-fold) and effective in working both with low-molecular and high-molecular substrates. Invention shows broad universality and provides applying biocatalysts in different reactors.
EFFECT: improved preparing method, improved, enhanced and valuable properties of biocatalyst.
