SUBSTANCE: invention proposes a novel synthetic gene encoding green fluorescent protein. The proposed gene is characterized by change of non-preferable or less preferable codons for preferable codons. The preferable codons are represented by the following group: GCC, CGC, GAC, TGC, CAG, GGC, CAC, ATC, CTG, AAG, CCC, TTC, AGC, ACC, TAC and AGG. Non-preferable codons represent the remained codons. The new synthetic gene is characterized by ability to express green fluorescent protein in the level 110-1 000% of the natural gene expression. Also, invention proposes a method for preparing the synthetic gene encoding green fluorescent protein. Method involves determination of non-preferable or less preferable codons in the natural gene and change one of some codons for preferable codons encoding the same amino acid as changing codons. Invention proposes also expressing vector comprising the synthetic gene and a method for preparing mammalian cultured cell line carrying the synthetic gene that involves the transformation of cells with expressing vector. The proposed invention can be used for industrial preparing the necessary proteins and for carrying out the genetic therapy also.
