FIELD: microbiology.

SUBSTANCE: invention relates, in particular, to a method for determination of type and level of hemolytic activity in B. anthracis. Method involves plating analyzed culture in spore or vegetative form on surface of bilayer blood agar with the following components, ml per Petri dish: backing agar, 15-20; cover agar, 5. Backing agar represents 1% agar "Difko" prepared in physiological solution. For preparing 100 ml of cover agar the following components are used: 50 ml beef-extract broth, 0.7 g of agar "Difko", 0.85 g of NaCl taking into account its content in beef-extract broth, and distilled water, up to 100 ml, followed by addition of 3-4 ml of ram erythrocytes suspension. Plating is incubated for 24-48 h. Type of hemolytic activity (α,β-hemolysis) is determined by visual method by pattern of color change of colony and hemolysis zone and also by ratio of colony radius (r_k) and hemolysis zone width and its level. The proposed method elicits high sensitivity and clearness.
