FIELD: medicine.
SUBSTANCE: method involves centrifuging supernatant at 14000 rpm during 5 min. Then, supernatant mixture with ZnSO4*7 H2O and 0.75N NaOH is shaken during 1 min and centrifuged at 14000 rpm during 5 min. Next to it, 100 ml of aliquot of Ph 7.5-8.0 is placed in chromatograph injector. Chromatographic analysis is carried out under the following conditions. Solid phase (chromatographic column) is prontosyl 120-5 C18 2*75 mm, dp=5 mc; movable phase (eluent) is 100% acetonitryl, phosphate buffer of pH=6.5. Chromatography mode of operation has ultraviolet absorption spectrum of 244, 262, 280 nm, time equal to 0.18 s, column temperature of 35°C. Antipyrin is determined from calibration plot showing chromatographic peak height dependency on preparation concentration.
EFFECT: high accuracy of the method; reduced number of working solutions; accelerated sample preparation; excluded toxic substances usage.
