SUBSTANCE: invention relates to a method for preparing cells used for cosmetic aims (for example, for rejuvenation and/or improvement of a patient skin state). In particular, invention concerns culturing autologous fibroblasts for their following using as an agent for substitution therapy. Method involves isolation of fibroblast cells from human skin bioptates by enzymatic treatment in solution of DMEM/5% EBS(embrional bovine serum)/0.2% dispase/0.1 mg/ml of collagenase of type I followed by centrifugation of cellular suspension, resuspending precipitated cells in medium DMEM/10% EBS/100 U/ml of penicillin, 100 U/ml of streptomycin, 100 U/ml of fungizone and prolonged culturing fibroblasts in medium with a patient serum: DMEM/10% MPS/100 U/ml of streptomycin, 100 U/ml of fungizone, or in therapeutic medium AIM-V. Invention allows averting the possible immune response and to obtain the better adaptability of cells in carrying out the substitution therapy.
