FIELD: chemistry, organic, genetic engineering.

SUBSTANCE: invention relates to genetic engineering of plants. Recombinant plasmid DNA pBi-P5CS will be constructed of the size 14681 bps providing transfer of target DNA sequence into the plant genome and lucerne P5CS gene expression consisting of the elements as follows: - DNA of the vector plasmid of the size 13915 bps; - DNA fragments containing kDNA of the gene Δ1-pyrroline-5-carboxylate synthetase of the size 2148 bps; - promoter 35S RNA from the genome of the cauliflower mosaics virus' genome; - 3'HTP of the nopalinsynthase gene area. The constructed recombinant plasmid pBi-P5CS carrying the gene Δ1-pyrroline-5-carboxylate synthetase will be transferred to the strain Agrohacterittm tumefaciens AGL0 with subsequent direct transformation of the Nicotiana tabacum SR1 leaf discs with cultivation together with agribacteria. Selection and generation will be performed directly on the media containing NaCl.

EFFECT: obtaining of stress-resistant tobacco plants with increased level of proline content using no antibiotics.
