FIELD: chemistry.
SUBSTANCE: isolated are neutrophils of peritoneal exudate of experimental animals, which are synthesized under impact of microbe cells Yersinia pestis EV - 76 general pool of neutrophilokins, from which fractions of neutrophilokins are isolated, from which fractions of neutrophilokins are isolated by precipitation with ammonium sulfate. Obtained sediment is separated by centrifuging with 8000 thousand rev/min, after which fractions are isolated by means of gel chromatography. Further, in phagocytosis reaction helper fractions of neutrophilokins are detected by adding to 1 ml of preliminary isolated macrophages of peritoneal exudate of animals 0.1 ml of suspension of plague microbe vaccine strain in amount 25 "м.кл" /macrophage and 0.1 ml fractions of neutrophilokins, containing 5-10 mkg of protein. Obtained suspension is mixed and underlaid under covering glasses, which are incubated, washed, fixed and stained after Romanovsky-Gimza. In stained test samples (with introduced neutrophilokins) and control samples (without fractions) percentage of active phagocytes (PAF) and index of phagocytosis completeness (IPC) are calculated, fractions, where IPC is [(+0.15±0.005)-(+0.68±0.04)], are isolated and defined as helper fractions. With introduction of helper fraction obtained according to claimed invention into animals, index of redistribution of subpopulations (IRS=21) grows in comparison with helper fraction of neutrophilokins, induced by latex microspheres (IRS=7.0), redistribution is towards quantitative increase of D subpopulations, the most important for formation of anti-plague immunity.
EFFECT: obtained helper fractions possess immunoregulating activity and can be used for correction of immunopathologic reactions in experimental plague.
