SUBSTANCE: offered way concerns medicine area, and 18 position 607 (C/A) concerns the method of determination of a genotype of the person on polymorphism in an interleukin gene. The way consists in use allele-specific primers with registration of PCR results in real time. Use primers, differing for everyone allele, the general for two allels primer and the general test of the following kind: Primer (allele C): 5' - GCG GCA GAA AGT GTA AAA ATT ATT AC - 3' Primer (allele A): 5' - GCG GCA GAA AGT GTA AAA ATT ATT AA - 3' the General primer (alleles C, A): 5' - CCT CCC AAG CTC AAT ATG GTG TC - 3 ' the General test (alleles C, A): (BHQ1) - 5' - TTG GTA TCC GTG TGG C (FdT)T GCA TCT GA-3' - (P) where BHQ1 - attached to 5'- tail nucleotide blank fluorescence damper, FdT - the fluorescent dye FAM attached to the nucleotide T.
EFFECT: revealing of a genotype of the person on polymorphism in an interleukin gene 18 position 607 (C/A), with reference of the investigated sample to a heterozygote or homozygote on one of the allele versions.
