SUBSTANCE: invention relates to molecular biology and biotechnology and may be used for development of DNA analysis systems. To determine, whether hot start takes place in polymerase chain reaction, according to the method, difference between PCR results for two reaction mixtures, which are the "hot start"-providing system under study and DNA matrix, is determined. PCR in one of the said reaction mixtures is carried out in the presence of oligonucleotide entirely complementary to 3'-terminal part of single-stranded DNA-matrix, and oligonucleotide identical to 5'-terminal part of single-stranded DNA-matrix. In the other reaction mixture, PCR is carried out in the presence of oligonucleotide partially non-complementary to 3'-terminal part of single-stranded DNA-matrix, and oligonucleotide identical to 5'-terminal part of single-stranded DNA-matrix.
EFFECT: possibility of reliable determination of "hot start", as well as of quality of the latter.
