SUBSTANCE: group of inventions relates to biotechnology, particularly to detection of a target nucleic acid sequence by a PCE-SH (PTO cleavage and extension-dependent signalling oligonucleotide hybridisation) assay. Present invention does not use probes to be hybridised with target nucleic acid sequences for providing target signals. Present invention uses probes (signalling oligonucleotides) to be hybridised with extended strand formed in a target-dependent manner in which extended strand is synthesised using templating oligonucleotide (CTO) artificially selected as templates.

EFFECT: disclosed is a method of detecting a target nucleic acid sequence by PCE-SH assay.
