FIELD: chemistry.
SUBSTANCE: invention relates to bioengineering and specifically to a method of purifying recombinant staphylokinase protein. The invention can be used in pharmaceutical industry. The method involves suspension of cells of the E. coli MZ08 strain - producer of recombinant staphylokinase, characterised by amino acid SEQ ID NO:1, in a phosphate buffer solution. The obtained supernatant undergoes homogenisation and centrifuging, followed by chromatography in two steps, the first comprising ion-exchange chromatography on a column filled with DE-cellulose to obtain a fraction rich in staphylokinase. The obtained fraction is then diluted with sodium chloride solution and the second step is carried out via metal-chelating affinity chromatography.
EFFECT: invention ensures high purity of recombinant staphylokinase protein.
