FIELD: bioengineering.
SUBSTANCE: invention relates to the method of producing of a recombinant protein SAV-RGD, where SAV is a streptavidin monomer, RGD is a melanoma addressing oligopeptide, having an amino-acid sequence Ser-Arg-Ala-Gly-Ala-Asp-Gly-Phe-Pro-Gly-Cys-Arg-Gly-Asp-Cys-Ser-Gln-Glu. The described method includes the cultivation in a culture medium of the bacteria strain E. coli MG1655/pSAV-RGD, produced by the transformation of the strain E. coli MG1655 by the plasmid pSAV-RGD, in its turn produced based on the vector pUC18 and including the sequence encoding the poured pro-protein SAV-RGD (pro-sav-rgd) under the control of the uridinephosphorylase promoter (Pudp) E. coli. The production of a fraction of cells of the bacteria strain, cleaning of the produced recombinant protein. Cultivation is carried out in two stages. At the first stage makeup by a glucose source is performed, and at the second stage makeup by peptone and yeastrel is performed.
EFFECT: increased efficiency of the use for the production of pharmaceuticals for the medical treatment of human melanoma.
