FIELD: medicine.
SUBSTANCE: what is described is an oligonucleotide hybridisation probe for B. mallei and B. pseudomallei identification by polymerase chain reaction by fluorescent detection. Fluorescent detection is implemented by means of a constructed oligonucleotide probe with complementary terminal "molecular beacon" sequences: 5'-(FAM)-CGCTGTCGACTTCGGCAACCAGCG-(RTQl)-3'. Where FAM is carboxyfluorescein, a fluorescent dye of absorption wave length 492 nm, and fluorescence wave length is 520 nm. RTQ1 is a fluorescence absorber with the absorbing range 470-570 nm. The use of the hybridisation probe provides high-sensitivity and specificity identification of glanders and melioidosis agents in a relatively short time in a biological material and environmental objects.
EFFECT: invention can be used in medicine for detection of a genetic material of glanders and melioidosis agents Burkholderia mallei and Burkholderia pseudomallei in samples both for diagnostics in practical public health service and Federal Service for Supervision of Consumer Rights and Human Welfare, and for scientific research.
