FIELD: medicine.
SUBSTANCE: invention describes creation of recombinant plasmid vectors pBI/rtTA/CITla and pBI/rtTA/Ltc3a, including minimal promoter of human cytomegalovirus, DNA sequence, coding antimicrobial peptides CIT1a and Ltc3a from Central Asian spider Lachesana tarabaevi, DNA sequence, coding transactivator protein for efficient expression of peptide genes. To inhibit Chlamydia trachomatis HEK293 cells are subjected to lipofection with obtained vectors, incubated in presence of doxycycline to induce expression of genes, coding antimicrobial peptides, after which cells are infected with Chlamydia. After incubation cells are stained and inclusions of Chlamydia are counted by means of monoclonal antibodies to superficial antigens of Chlamydia, conjugated with fluoroscein. Level of inhibition is determined by reduction of number of Chlamydia inclusions in transformed cells in comparison with control - number of Chlamydia inclusions in non-transformed cells.
EFFECT: method ensures required level of chlamydial infection inhibition.
