SUBSTANCE: invention is a recombinant plasmid DNA pHIAsp03, directing the synthesis of human hybrid protein with human proinsulin Aspart containing DNA encoding the hybrid protein with human proinsulin Aspart with size of 134 amino-acid residue with the amino acid sequence shown in Figure 2, comprising the amino acid sequence of the leader peptide in the form of N-terminal fragment of human gamma-interferon, connected to proinsulin Aspart with the peptide linker. On the basis of the recombinant plasmid pHIAsp03 the strain Escherichia coli JM109/pHIAsp03 was obtained - producer of the hybrid protein with proinsulin Aspart, registered in the federal state unitary enterprise State Research Institute of Genetics RNCIM under the number B-11411.
EFFECT: use of the claimed inventions enables to simplify the technology of isolation of insulin Aspart and to increase its yield.
