FIELD: chemistry.
SUBSTANCE: group of inventions relates to the field of microbiology and biotechnology. A method of detecting alive cells of a microorganism in a tested sample by differentiation of the alive cells from the dead cells or injured cells includes addition of a preparation, capable of covalent binding with DNA or RNA when irradiated by light with a wavelength from 350 nm to 700 nm, into the tested sample, irradiation of the tested sample; amplification of the target area of DNA or RNA of the microorganism, contained in the tested sample, by a method of amplification of nucleic acids in the presence of a preparation suppressing the action of a substance, which inhibits amplification of nucleic acids, magnesium salt, salt of organic acid or salt of phosphoric acid, without isolation of nucleic acids from the cells and analysis of the amplification product. The method can be realised by an application of a kit, consisting of a preparation, capable of covalent binding with DNA or RNA when irradiated by light with a wavelength from 350 nm to 700 nm, a preparation, suppressing the action of a substance, inhibiting amplification of nucleic acids, and primers for amplification of the target area.
EFFECT: claimed inventions make it possible to detect the alive cells of microorganisms in various biological samples with high sensitivity and accuracy.
