FIELD: chemistry.
SUBSTANCE: invention relates to biotechnology. Disclosed is a method of breaking down ribonucleic acids. A solution containing ribonucleic acid is passed through a macroporous monolithic polymethacrylate sorbent containing immobilised ribonuclease A and then through a macroporous monolithic anion-exchange sorbent. The following elution program is used with mobile phase velocity of 10 cm/min: the first step includes using a solution A, which is 0.02 M tris-HCl buffer solution, pH 7.5; the second step includes using a solution B, which is solution A containing 7 mol/l urea, as well as linear gradient elution of 0-100% of solution C, which is solution B containing 1 mol/l sodium chloride.
EFFECT: advantage of the invention is simultaneously performing efficient RNA breakdown and monitoring reaction products over a short period of time with high mobile phase velocities and low operating pressure.
