SUBSTANCE: invention relates to a method of determining the binding constant of a high-affinity compound with proteins, which includes the following steps: a) adding a high-affinity compound to a two-chamber system and determining the amount of the high-affinity compound in one of the chambers after reaching equilibrium distribution, b) adding an affinity reducing compound to one of the chambers and determining the distribution coefficient of the high-affinity compound on the affinity reducing compound after reaching equilibrium distribution, c) adding a nonspecific protein to the other chamber and determining the distribution coefficient of the high-affinity compound on the nonspecific protein in the presence of the affinity reducing compound after reaching equilibrium distribution, and d) determining the binding constant of the high-affinity compound with the distribution coefficient from steps b) and c).

EFFECT: invention is efficient when searching for a substance with the best physical and chemical properties and for understanding the relationship between a chemical structure and physical and chemical parameters.
