FIELD: chemistry.
SUBSTANCE: group of inventions relates to biotechnology and specifically to a method of cultivating a single B-cell, which is obtained from a population of B-cells of an experimental animal. The method includes co-cultivating each B-cell with murine EL-4 B5 cells as feeder cells and selecting B-cell clone secreting an antibody, followed by co-cultivation in the presence of a culture medium containing IL-1β, TNFα, IL-10 and one or more components selected from IL-21, Staphylococcus aureus strain Cowans cells (SAC), B-cell activation factor of the tumour necrosis factor family (BAFF), IL-2, IL-4, IL-6. That way, B-cells proliferating and secreting IgG are selected. Labelled IgG+CD19+-, IgG+CD38+-, IgG+CD268+-, IgG-CD138+-, CD27+CD138+- and CD3-CD27+-B-cells are also obtained using surface markers.
EFFECT: group of inventions enables to obtain induced B-cells producing antibodies and to determine binding specificity thereof.
