FIELD: chemistry.
SUBSTANCE: group of inventions relates to method for amplification of genome locus in bacterial cell in vivo, bacterial host cell, containing it and capable of its amplification, as well as to culture of bacterial cells, capable of genome locus amplification. Method includes introduction of locus with structure A1-P-M-A2 into population of host cells, bringing said cell population in contact with inhibitor of enzyme, required for cell viability, cultivation and selection of cells, resistant to said inhibitor. In locus structure A1 and A2 are direct repeats, P contains coding sequence for protein of interest, capable of expression in bacteria, M contains coding sequence for enzyme, functionally bound with promoter. Cells, which are resistant to said inhibitor, have multitude of copies of amplification unit.
EFFECT: group of inventions provides increased level of expression of protein of interest in host cells.
