METHOD OF PRODUCING PENTAHYDROXYETHYLNAPHTHOQUINONE (ECHINOCHROME A) Russian patent published in 2016 - IPC C07C46/00 C07C50/32 

FIELD: chemistry; pharmaceutics.

SUBSTANCE: invention relates to a method for producing pentahydroxyethylnaphthoquinone (echinochrome A), which is active substance for manufacture of medicaments: Histochrome® solution for injections of 0.2 mg/ml and Histochrome® solution for intravenous administration of 10 mg/ml and for production of biologically active food additives, from sea urchin spawn. Method consists in that freshly caught or defrosted sea urchins are extracted with 96 % ethanol with addition of a mineral acid or acetic acid in ratio of raw materials: extractant of 1:(5-10) for 6-10 hours at room temperature, then resulting extract is concentrated under vacuum and extracted twice with chloroform in a volume ratio chloroform: water of 1:(1-2); further chloroform extracts are combined and concentrated under vacuum until complete removal of solvent, and then applied as a dry charge to chromatographic column packed with sorbent polychrome-1, column is washed further with distilled water and product is eluted with 25-35 % ethyl alcohol solution; further eluate was concentrated in a vacuum and re-chromatographed on a column packed with sorbent Toyopearl HW-40 or Toyopearl HW-50, 40 % ethanol with 0.1 % formic acid, resulting eluate is concentrated to dryness, desired product is crystallised from ethyl alcohol or acetone, dried and sublimated.

EFFECT: proposed method allows to produce end product with high output and high degree of purity.
