SUBSTANCE: invention is related to phthisiology, and can be used for detection of tuberculosis causative agent in microbiological cultures. For this purpose, the diagnostic technique for mycobacterium tuberculosis includes sandwich immunoassay with the use of the primary and secondary antibodies directed against a secretory protein MRV64 (24 kDa) specific for Mycobacterium tuberculosis complex. Two monoclonal antibodies directed against two different mycobacterial protein MPT64 (24 kDa) epitopes, possessing specificity and high affinity toward analysed antigen and having different isotypic appurtenant are used. At that, one monoclonal antibody is fixed to the solid phase, test material is added and incubated after washing of non-adsorbed antibodies. During the formation of the antigen MPT64 (24 kDa) - monoclonal antibody complex another monoclonal antibody is added and bound to the complex after the next washing, then it is recognised by peroxidase labeled isotype-specific antibodies for detecting the complex in a quantitative manner based on standards with known protein MPT64 (24 kDa) concentration.
EFFECT: using this technique enables to specifically detect the presence of Mycobacterium tuberculosis with the help of two monoclonal antibodies directed against two different Mycobacterial protein MRT64 epitopes.
