FIELD: chemistry.
SUBSTANCE: invention relates to analytical chemistry, specifically to determination of malonic dialdehyde in vegetative organs of sweet cherries, grapes, walnut and winter wheat seedlings. Method comprises extraction of biological material with 96 % alcohol and with subsequent centrifugation. Further, supernatant is treated with 0.5 % aqueous solution of thiobarbituric acid and incubated for 30 minutes at 50 °C. After cooling reaction mixture is centrifuged. Malonic dialdehyde is determined by capillary electrophoresis in a quartz capillary with inner diameter of 75 mcm and effective length of 0.5 m. Analysis is performed at wavelength 254 nm in a stream of electrolyte, containing 0.33 wt% boric acid and 0.05 wt% sodium tetraborate at positive voltage polarity. Calculation is carried out taking into account that two molecules of thiobarbituric acid are used per molecule of malonic aldehyde.
EFFECT: invention enables to conduct quantitative determination of concentration of malonic dialdehyde, which serves as a marker of oxidative stress in plants.
