FIELD: biotechnology.
SUBSTANCE: invention relates to biotechnology and represents a method for site-specific hydrolysis of C5-methylated DNA sequence. Reaction mixture is prepared containing a buffer solution and a sample of C5-methylated DNA, the mixture is added with dimethyl sulfoxide to the final concentration of 15-25 %, and then with MD-endonuclease, the mixture is incubated for one hour at 30-37 °C, followed by analysis of the DNA hydrolysis result by gel electrophoresis, herewith the MD-endonuclease used is GlaI with the concentration of 4-8 ea/mcl or PcsI with the concentration of 1-2 ea/mcl.
EFFECT: invention provides higher efficiency of site-specific hydrolysis of C5-methylated DNA sequence.
