FIELD: biotechnology.
SUBSTANCE: invention represents recombinant bacterial strain Escherichia coli N42 (pElmI) being a producer of methyl-dependant site-specific DNA-endonuclease ElmI splitting both chains of DNA sequence 5′-GC^NGC-3′/3′-CGN^CG-5′, in which at least three cytosines are methylated in the C5 position to form 5′-projecting nucleotide ends. Strain is produced by cells transformation of Escherichia coli ER2267 with pElmI plasmid. This plasmid is designed on the basis of vector pMTL22 and contains a gene coding the methyl-dependant site-specific DNA-endonuclease ElmI from the natural strain Escherichia coli N17.
EFFECT: invention enables to obtain DNA-endonucleases ElmI of a specified specificity with higher output.
