FIELD: biotechnology.
SUBSTANCE: invention relates to biotechnology. Described is a method of producing mature megakaryocyte cells. Method involves forced expression o oncogene selected from MYC genes and a gene selected from BMI1, Ring1a, Ring1b, Phc1, Phc2, Phc3, Cbx2, Cbx4, Cbx6, Cbx7 and Cbx8, in haemopoietic precursor cells, CD34-positive cells or megakaryocyte precursor cells without multi-polyploidisation, and cultivation and growing of said cells; and differentiation of grown cells into mature megakaryocyte cells. Also described is a method for producing a preparation of thrombocytes, involving: obtaining mature megakaryocyte cells using described method; extraction of culture of megakaryocyte cells of a fraction of a cultural solution which rich with thrombocytes, released from megakaryocyte cells; and removal from said fraction of other components of blood cells different from thrombocytes. Disclosed are cell preparations for producing mature megakaryocyte cells and thrombocytes, containing megakaryocyte precursor cells without multi-polyploidisation or mature megakaryocyte cells, into which is introduced an oncogene, selected from MYC genes and a gene selected from genes BMI1, Ring1a, Ring1b, Phc1, Phc2, Phc3, Cbx2, Cbx4, Cbx6, Cbx7 and Cbx8.
EFFECT: described are kits for producing megakaryocyte precursor cells at a pre-multinucleation step, for producing mature megakaryocyte cells for producing thrombocytes.
