FIELD: biochemistry.

SUBSTANCE: invention relates to biochemistry, biotechnology and medicine, namely to method of producing recombinant anti-tumor toxin-chimeric bifunctional protein, consisting of targeted polypeptide Darpin 9-29, specific to histochemical marker HER2/neu, and highly active bacterial ribonuclease barnase, connected with flexible peptide linker. For implementation of this method recombinant strain is produced Escherichia coli BL21(DE3)/pETDa by embedding into strain of E.coli BL21(DE3) plasmid pETDa. Said plasmid is produced by cloning of recombinant DNA fragment, coding said chimeric protein, in plasmid vector pET22 at restriction sites NdeI/HindIII.

EFFECT: present invention enables to obtain targeted protein characterized by low risk of side effects due to instant cancellation of cytotoxicity for targeting on HER2/neu therapy of tumors overexpressing HER2 receptor.
