FIELD: biotechnology.
SUBSTANCE: invention can be used to obtain recombinant allergen of Pisum sativum field pea. The plasmid vector pET-pPsLTP is constructed for expression of Pis s 3 in cells of Escherichia coli in the composition of the hybrid protein His8-TrxL-Pis s 3. The parent Escherichia coli BL21 (DE3) Star strain is transformed by the proposed plasmid vector obtaining a producer strain of hybrid protein His8-TrxL-Pis s 3. Then the cultivation and subsequent cell lysis of the producer strain are done. The affine purification of the hybrid protein His8-TrxL-Pis s 3 is performed on a metallochelated carrier. The hybrid protein His8-TrxL-Pis s 3 is split by bromine cyan in the methionine residue inserted between sequences of allergen Pis s 3 and thioredoxin. The metallochelated chromatography is repeated and the target peptide purification is carried out by using reversed phase HPLC.
EFFECT: invention allows for obtaining highly purified food allergen Pis s 3 able to bind specific antibodies.
