SUBSTANCE: method involves cultivation of the E. coli strain B121 (DE3)/pEGT1/H1.3S producing the target protein, biomass disintegration, target protein extraction with perchloric acid, and subsequent purification of the target protein. This method is characterized by protein purification involving purification by cation exchange chromatography using washing of a sorbent coated with protein with a solution of 4 M urea and purification by HPLC. This method also comprises concentration of the purified bismethionylhistone by ultrafiltration and lyophilization to obtain a purified bismethionylhistone powder with a purity of 98.5%.
