FIELD: biotechnology.
SUBSTANCE: method for producing a molecular complex, having a GAA (acid alpha-glucosidase) activity, including the contact of a polypeptide with GAA activity with protease of Aspergillus oryzae fungus. Said protease macerates said polypeptide at one or more sites between amino acid 50 and amino acid 74 of the sequence of said polypeptide. An isolated fungal cell is presented. It comprises of a nucleic acid encoding the amino acid sequence of said GAA polypeptide and a nucleic acid encoding said alkaline protease. Said fungal cell produces said molecular complex having GAA activity and comprising of at least two polypeptides.
EFFECT: group of inventions allows for the production of more mature forms of the GAA polypeptide, which have an increased specific activity and improved ability to reduce glycogen levels in comparison with the GAA precursor and the commercial GAA of a human.
