SUBSTANCE: invention relates to field of biotechnology, namely to method for reducing level of O-glycosylation of insulin precursor molecules or insulin analogues, synthesised by Pichia sp. Invention discloses knockout strains of methylotrophic yeasts, obtained by means of genetic engineering. Strains contain disruption in protein-mannosyl transferase (PMT) genes and are capable of synthesising heterologous proteins with reduced degree of glycosylation. Also described is vector, containing coding sequences of PMT1, PMT2, PMT4, PMT5 and PMT6 genes for transformation of methylotrophic yeasts.
