FIELD: medicine.
SUBSTANCE: invention relates to medicine. Invention is a set of specific oligonucleotide primers for the Acinetobacter genus bacteria species identification and the resistance to beta-lactam antibiotics determination, comprising: 2 pairs of oligonucleotide primers for the rpoB gene amlification, with nucleotide sequences atgcgtccaggcgagccaccaac, gcggctgaaa acttattcaa taac, tctg aacgctatga cttatc, atggtcgtgtttgtccaattgaa; 3 pairs of oligonucleotide primers for the OXA-type carbapenemases genes amlification (oxa-51 and oxa-51-like), with nucleotide sequences gcaccataaggcaaccaccaca, aagtatttaaatgggatggg, agaatgggaaaagaacatgacc, ttggacttgagctcatgtctaag, gcaccataaggcaaccacccgg, aagtatttaaatgggatgcc; 2 pairs of oligonucleotide primers for the oxa-23 carbapenemase gene amlification, with nucleotide sequences atctggtgtttaaaatgaataa, gattcatcaatactttgatgaa, tggaagggcgagaaaaggtcat, ggcaagggatataaaaccacaa; 2 pairs of oligonucleotide primers for the oxa-40 carbapenemase gene amlification, with nucleotide sequences tgaagctcaaacacagggtgta, taaaaaaagaacttatcctatgtg, ccagtatatcaagagcttgcaag, ccttaaaattacaccagtacaagaag. Method for Acinetobacter bacteria identifying and determining their resistance to beta-lactam antibiotics involves the DNA isolation from the bacterial cultures bio-material samples and carrying out PCR with synthetic oligonucleotide primers, using the proposed set of oligonucleotide primers, and in the case of rpoB and oha-51 genes detection, bacteria of the Acinetobacter genus are identified, and in the presence of ocho-23 and oha-40 genes encoding carbapenemases OXA-23 and OXA-40, conclude that the analyzed strain is resistant to antibacterial drugs of the beta-lactam number.
EFFECT: invention makes it possible to identify Acinetobacter bacteria directly in a clinical material with simultaneous determination of resistance to OXA type carbapenems; and can also be used in the selection of a drug, treatment regimen and for epidemiological control.
