FIELD: biotechnology.
SUBSTANCE: method for making a system of delivery of nucleic acid fragments (NAF) to mammalian cells is provided. The base for the delivery of NAF is synthesized. Amine-substituted silanol (Si~NH2) at a concentration of 1-4 mg Si/ml is used as the base. Si~NH2 is obtained by hydrolysis in water of aminopropyltriethoxysilane at a pH value of 10-11 and temperature of 20-70°C followed by neutralisation of silanol with a 0.5-1% solution of acetic acid to a pH value of 8-9. The resulting solution of Si~NH2 is added with the calculated number of 10-3-10-7 M of the corresponding NAF solution in the ratio of 5-100 nmol of NAF per 1 mg of Si and incubated in 0.1-0.2 M of NaCl at room temperature for 20-30 min. A system of delivery of NAF to cells is made. It is a composite of Si~NH2⋅NAF with a NAF capacity of 10-100 nmol/mg of Si.
EFFECT: high strength of binding of oligonucleotide with the base, ability to penetrate through the cell membrane and low cytotoxicity.
