FIELD: biotechnology.
SUBSTANCE: invention relates to biochemistry and biotechnology, in particular to a method for creating and selecting a library of modify aptamers for various targets, in particular to the human cellular receptor CD47. Represented method is characterized in that, that synthesize combinatorial libraries of oligonucleotides consisting of constant sites, necessary for amplification of library fragments in selection process, and a variable region comprising modified by reaction azide-alkene coupling nucleotides, which are non-canonical nucleotides, such as 5'-substituted deoxy-uridines, are incubated with a target protein and affinity variants of aptamers are selected, followed by amplification using non-canonical minimally modified nucleotides, followed by carrying out the azide-alkyne coupling reaction to obtain modifications, presented in the source library, and further selection of affine target-specific molecules.
EFFECT: method allows to select quickly complexly modified aptamers to different targets with increased binding potential.
