METHOD OF SELECTING SINGLE-STRANDED OLIGONUCLEOTIDES WITH HIGH AFFINITY TO MOLECULAR TARGET Russian patent published in 2020 - IPC C12N15/00

Abstract RU 2736789 C1

FIELD: biotechnology.

SUBSTANCE: invention relates to biotechnology and can be used in selection from combinatorial libraries of individual single-chain oligonucleotides or families of oligonucleotides with required functions. To impart increased structural diversity and high affinity to the target, the oligonucleotides contain up to 4 % modified heterocyclic bases. To obtain oligonucleotides, a low concentration of combinatorial libraries is used, which is individual for each round of selection, which enables to control complexation between oligonucleotides and a molecular target and neutralize the effect of non-target and nonspecific interactions with a carrier used for immobilising a molecular target. Described also is plant for selection and necessary preparatory experiments.

EFFECT: resulting oligonucleotides are characterized by low dissociation rates and can be used as analogues of monoclonal antibodies in typical application scenarios.
