FIELD: medicine.

SUBSTANCE: invention relates to medicine, namely to laboratory diagnostics, and can be used to prepare erythrocytes in a fluorescent in situ hybridization (FISH) method. To do this, the patient's blood is washed three times by centrifugation at 1,000 rpm in phosphate-buffered saline at pH 7.4 for 5 minutes. It is then adjusted to a one millionth cell concentration in a milliliter. After that a 1 ml sample is pelleted by centrifugation at 1,000 rpm for 5 minutes. Resuspend in 400 mcl 0.5 % glutaraldehyde solution and fix for 30 min at 25 °C. Fixed erythrocytes are washed with phosphate buffered saline at pH 7.4. Then again resuspended in ethanol solutions with an ascending concentration 50, 80 and 100 % by 400 mcl of each solution, followed by incubation for 10 min at 4 °C. Red blood cells are then rinsed in phosphate buffered saline with pH of 7.4 by centrifugation at 1,000 rpm for 5 minutes and resuspended in 300 mcl of distilled water.

EFFECT: invention allows to isolate erythrocytes from the test blood samples without changing their size and shape while maintaining clearly defined cell boundaries and central cavity of the disc and to reveal bacteria on the surface and inside the red blood cells during the diagnosis of septic states of patients.
