SUBSTANCE: invention relates to biotechnology. Described is a method for T-cell activation assay by detecting copies of at least one RNA molecule expressed in individual T-cells, comprising providing a sample containing a population of cells, containing T-cells; inducing gene expression in T-cells ex vivo, either immediately or after culturing, by incubating cells with at least one compound that binds to specific receptors and initiates gene expression; fixation and permeabilization of cells; labelling copies of at least one RNA molecule expressed by T-cells using a set of fluorescently labelled oligonucleotide hybridisation probes that have complementary sequences to RNA molecules and removal of unbound probes by washing; and detection of cells with expression events by flow cytometry (FC), where the expression event is one or more fluorescence measurements that are grouped by gating fluorescence intensity.
EFFECT: invention makes it possible to quickly detect copies of at least one RNA molecule expressed in individual cells.
