FIELD: biochemistry.
SUBSTANCE: invention relates to the field of biochemistry and molecular biology. Method is proposed for determining the share of mtDNA with deletions in biological samples, including the isolation of DNA from samples, simultaneous amplification of the corresponding DNA segments in triplex mode using PCR in real time with the subsequent determination of the starting quantities of selected DNA fragments, characterized in that for each sample simultaneously amplify and detect D-loop mtDNA fragments and ND4 and nuclear DNA LINE-1 using the FAM, HEX, ROX channels, determine the starting quantities of these DNA fragments, next, the relative amount of mtDNA is calculated by the ratio of [D-loop]/[LINE-l], after which the proportion of mtDNA is determined with deletions from the total amount of mtDNA using the formula l-[ND4]/[D-loop].
EFFECT: this method allows reducing errors in detecting mtDNA deletions.
