SUBSTANCE: invention relates to molecular biology. Described is a method of producing a full-length sequence of human mitochondrial DNA to identify an unknown individual and determine biological relationship by multiplex amplification for working with degraded DNA samples. Method is characterized by that for genotyping of mitochondrial DNA, composition of 446 original synthetic oligonucleotide primers is used for conducting multiplex polymerase chain reaction, flanking DNA sections containing short overlapping sections of human mitochondrial DNA.
EFFECT: selection of synthetic primers, which enable to amplify short overlapping sections of human mitochondrial DNA for subsequent sequencing using a method of next-generation sequencing (NGS), or by Sanger's method, or other sequencing methods, with subsequent alignment of readings on reference mitochondrial genomes of individuals from Russian populations.
