HYBRID PROTEINS ALIKE ALKALINE PHOSPHATASE Russian patent published in 2019 - IPC C12N9/16 C12N15/55 A61K38/46

Abstract RU 2683635 C2

FIELD: biotechnology.

SUBSTANCE: group of inventions relates to biotechnology. Proposed isolated protein with phosphatase activity, containing the amino acid sequence of at least 365 consecutive amino acids, having at least 98 % identity with SEQ ID NO: 5, the amino acid sequence of 65 consecutive amino acids, having at least 98 % identity with SEQ ID NO: 6, and an amino acid sequence of 54 consecutive amino acids having at least 98 % identity with SEQ ID NO: 7, where the full-size protein contains an amino acid sequence having at least 98 % identity with the full amino acid sequence of SEQ ID NO: 1, when it contains modifications 279L, 328V and 478L. Polynucleotide encoding the specified protein, and a vector containing the specified polynucleotide is proposed. Proposed host cell containing the specified protein, polynucleotide or vector, is a cell CHO. Method is proposed for producing said protein using said host cell. Proposed pharmaceutical composition contains the specified protein for use as a drug for the prevention or treatment of inflammatory diseases, kidney disease or hypophosphatasia.

EFFECT: group of inventions allows to obtain a protein with phosphatase activity, with improved thermal stability, improved biodistribution and improved Zn2+ independence and which may be useful for treating diseases caused by or accompanied by low levels of Zn2+.
