FIELD: medicine.
SUBSTANCE: invention relates to laboratory diagnostics, and can be used for selection of optimal cryoprotector by content of glycogen in leukocytes of preserved blood. That is ensured by obtaining a portion of venous blood before the onset of cryogemotransfusion therapy, dividing it into equal parts into test tubes, in one of which the control sample (CS) of blood does not contain a cryoprotector, while another sample is added with one equal dose of tested cryoprotectors - test samples (TS). Tubes are sealed with plugs, the contents are mixed on a shaker at a temperature of plus 37 °C for 4 hours, drops of TS and CS are applied in volume of 4 mcl on the slide, 2-3 smears are made, quickly dried in air at room temperature, is fixed in 10 % of alcohol-formalin mixture, stained for glycogen by method of Shabadash, leukocytes are microscoped in transmitted light. Comparative study of the average cytochemical coefficient of segment-nucleated (c/i) neutrophils and lymphocytes of TS and CS; if the values of the TS cytochemical coefficient and the CC cytochemical coefficient are equal, the tested cryoprotector is evaluated as optimal for the particular patient and suitable for use; at TS cytochemical coefficient values, other than CS cytochemical coefficient, conclusion is made as unfit to cryoprotector.
EFFECT: invention enables individual selection of an optimal cryoprotector at the preclinical stage of cryogemotransfusion therapy.
