FIELD: medicine; biochemistry.
SUBSTANCE: invention relates to a method for production of collagenous osteoplastic material from bone tissue, consisting in the fact that after mechanical cleaning and fragmentation material is washed up heated to 45–50 °C saline solution for 30 minutes is treated with solution of 3 % hydrogen peroxide in water in form of 3 cycles for 10 minutes with washing with saline solution at 45–50 °C, and instead of papain treatment, treating with 0.1–0.5 % lipase in buffered physiologic saline at pH 8.0 for 2 days, then washed with physiologic saline, treated with a mixture of chloroform with water (1:1) for 1–3 days with intermittent replacement of the mixture as it becomes turbid, chloroform is washed with solution of 20–30 % ethanol in water for 6–12 hours, and then with physiological solution at fivefold change of solutions, material is demineralised with solution of 0.6 M hydrochloric acid in water for 0.25–1 hours at temperature 4–6 °C under conditions of mixing and changing solution to fresh solution every 15 minutes, then treated with 5 % sodium thiosulphate solution in water for 3 hours, washed with 10-fold change of distilled water, and normalized in buffered physiological solution of pH 7.4 for 7 days at room temperature and with change of solution to fresh 3 times a day, wherein ratio of volumes of bone tissue and mixtures or solutions for treatment or washing is 1:4–1:6, treatment and washing are performed in conditions of mixing at 40–50 °C, unless other temperature is specified, and demineralisation is performed at periodic evacuation to 10–30 mm Hg and gas phase pressure normalization over material with period of 2–10 minutes.
EFFECT: method enables to obtain material with high degree of preservation of collagen matrix structure, biocompatibility, bio-integration, and osteoplastic potential.
