FIELD: biotechnology.
SUBSTANCE: invention represents a method for obtaining an antigen from Mycobacterium bovis Bovinus-8 strain 700201 with molecular weight of 28 kDa for studying the humoral immune response, involving cultivation of an agent on a solid nutrient medium of Levenshtein-Jensen medium, destruction of washed cells and fractionation by preparative electrophoresis, where washed cells are subjected to destruction three times in homogenizer Fast Prep 24 for 45 sec at vibration rate of 6.5 ms, destructed cells are removed by centrifugation at 7,000 rpm, maintained supernatant at +4–6 °C for 10 days, repeatedly removing spontaneous sediment by centrifugation, adding to supernatant lysing buffer containing 0.2 % sodium dodecyl sulphate, 0.1 % 2-β mercaptoethanol, samples are held at 100 °C from 10 minutes, fractionated on column using 9 % polyacryamide gel.
EFFECT: invention enables to obtain an antigen having high specificity and activity.
