FIELD: medicine.
SUBSTANCE: invention refers to medicine, particularly to the determination of the drug metformin in the mixed saliva of a patient suffering diabetes mellitus. For this purpose, 100 mcl of intact saliva is transferred into 1.2 ml microfluid tubes, 10 mcl of internal standard solution is added in concentration of 1,000 ng/ml, then it is mixed on shaker at 1,200 rpm for 2 minutes, after which proteins are precipitated by addition of 300 mcl cooled down to -20 °C acetonitrile, then shaking at 1,200 rpm for 4 minutes, obtained mixture is centrifuged at 4,000 rpm at temperature +4 °C for 15 minutes, after that supernatant fluid in amount of 50 mcl is mixed with 150 mcl of water and placed into plates for determining concentration of metformin by high-performance liquid chromatography, wherein linear range of determining drug metformin ranges from 5 to 1,600 ng/ml.
EFFECT: invention provides simplifying the process of detecting the drug metformin, high accuracy of diagnosing the toxic effect of the drug metformin.
