FIELD: biotechnology.
SUBSTANCE: group of inventions relates to biotechnology. Polypeptide having O-acetylhomoserine release activity, wherein at least one amino acid selected from a group consisting of phenylalanine in position 30, leucine in position 95 and phenylalanine in position 165 of amino acid sequence SEQ ID NO: 1, is replaced by another amino acid, and optionally valine in position 1 of amino acid sequence SEQ ID NO: 1 is additionally replaced by methionine. Polynucleotide encoding said polypeptide. Escherichia genus microorganism producing O-acetyl-homoserine containing or overexpressing the modified polypeptide of the amino acid sequence SEQ ID NO: 1, having a substitution of at least one amino acid selected from a group consisting of phenylalanine in position 30, leucine in position 95 and phenylalanine at position 165 of amino acid sequence SEQ ID NO: 1, and where, optionally, in said modified polypeptide, valine in position 1 of amino acid sequence SEQ ID NO: 1 is additionally replaced by methionine. Method of producing O-acetyl-homoserine involves culturing said Escherichia genus producing O-acetyl-homoserine in a medium and recovering O-acetyl-homoserine from a cultured microorganism or culture medium. Method of producing L-methionine involves culturing said Escherichia genus producing O-acetyl-homoserine in a medium and converting O-acetyl-homoserine to L-methionine by treating said cultured microorganism or culture medium or O-acetyl-homoserine recovered from a cultured microorganism or from a culture medium, methylmercaptan and methionine-converting enzyme.
EFFECT: group of inventions provides producing protein capable of releasing O-acetyl homoserine.
