FIELD: biotechnology.
SUBSTANCE: invention relates to the field of biotechnology. Invention is a method for cultivation of callus tissue Vaccinium myrtillus L. – producer of phenol compounds, and can be used in medicine for production of raw materials, rich in flavonoids, regardless of season, during cultivation of callus tissues of other slowly growing plants, for cell selection, genetic transformation, fundamental physiological-biochemical studies. Method involves preparation of Woody Plant Medium nutrient medium while heating. Additionally hormones are added to nutrient medium: α-naphthyl acetic acid – 0.5 mg/l and 6-benzylaminopurine – 0.5 mg/l. Obtained nutrient medium is poured on culture vessels, closed and autoclaved. To produce callus, leaves of sterile plants Vaccinium myrtillus L. are incised with a scalpel, leaf explants are placed in culture vessels with frozen nutrient medium, sterile closed and stored on shelves with illumination. Produced callus tissues are separated from the nutrient medium, cut into parts and transferred to the fresh nutrient medium of the same composition. Subculture of callus is carried out every 4–6 weeks with 16-hour photoperiod.
EFFECT: invention increases accumulation of biomass and phenol compounds, including flavonoids, in callus tissue Vaccinium myrtillus L.
