SUBSTANCE: invention relates to biochemistry. Application is described in vitro modified lymphocytes containing a vector for controlled expression of protein having function of interleukin-12 (IL-12) for treating a tumour in a mammal, wherein said vector comprises a polynucleotide encoding a gene switch, wherein said gene switch comprises: (a) at least one transcription factor sequence, wherein said at least one transcription factor sequence encodes a ligand-dependent transcription factor operably linked to a promoter; and (b) a polynucleotide encoding a protein having an IL-12 function associated with a promoter which is activated by said ligand-dependent transcription factor; wherein said polynucleotide encoding said protein, having IL-12 function, encodes a protein which is at least 85 % identical to human wild-type IL-12, and wherein said ligand is administered in an effective amount in, or less than 24 hours after intra-tumour delivery of an effective amount of said lymphocytes.
