SUBSTANCE: invention relates to biotechnology and immunology. Disclosed is a method of producing a human IgG Fc region containing a first variant polypeptide of the Fc region and a second variant polypeptide of the Fc region. First variant polypeptide of the Fc-region is derived from the first parent polypeptide of the Fc region of the IgG class, and the second variant polypeptide of the Fc-region comes from the second parent polypeptide of the Fc-region of the IgG class. First parent polypeptide is identical to the second parent polypeptide or differs from the second parent polypeptide. Mutations are introduced into parental polypeptides, including a combination of: T307H and N434H, where the numbering corresponds to the EU index by Kabat. As a result, the first variant polypeptide of the Fc region differs from the second variant polypeptide of the Fc region by one or more amino acid residues. If the first parent polypeptide is different from the second parent polypeptide, the first variant polypeptide differs from the second variant polypeptide by residues differing from those amino acid residues from which the first parent polypeptide differed from the second parent polypeptide.

EFFECT: obtained human IgG Fc-region has an affinity for the human neonatal Fc receptor, which is higher than that in the Fc region of the IgG class without mutations, which can be used in treating diseases requiring an extended retention time of the antibodies.
