FIELD: biotechnology.
SUBSTANCE: method of DNA determination in biological fluids, which consists in preparation of nanoparticles of noble metals acting as a GRS-amplifier; target hybridisation with a targeted covalently GRS-labeled complementary oligonucleotide; applying a sample with the analyzed compound to the surface of liquid sample nanoparticles with the analyzed compound to form a structure directed structurally by non-covalent interactions, including a DNA to be determined, a GRS mark and an oligonucleotide; the obtained hybridised GRS-labeled duplex is detected by giant Raman scattering spectroscopy. Qualitative and quantitative DNA content is determined from the position and intensity of the bands on the recorded spectra. Method is applicable for detecting small amounts of DNA against the backdrop of a large amount of foreign components of the analyzed biological fluids.
EFFECT: high sensitivity, selectivity and rapidness.
